BACKGROUND
Tumor necrosis factor-alpha (TNF-α, also known as TNF-alpha, TNF-a, TNFa, Tumor necrosis factor-alpha, cachexin, cachectin or TNFSF2) is a cell signaling protein (cytokine) involved in systemic inflammation and is one of the cytokines that make up the acute phase reaction.
It is produced chiefly by activated macrophages, although it can be produced by many other cell types such as CD4+ lymphocytes, NK cells, neutrophils, mast cells, eosinophils, and neurons. TNFα is a member of the TNF superfamily, consisting of various transmembrane proteins with a homologous TNF domain
TNF alpha plays a proinflammatory role during a tissue injury or a lesion, sometimes by himself or with mediators like interleukins 1 and 6.
TNF alpha has a cellular intervention where it uses lymphocytes and neutrophils but also permits tissues to remodel and repair.
TNF alpha has a positive action and will protect the body if the immune system is functioning properly. However, if the Tnf-alpha levels are altered it can lead to serious pathologies such as Behçet decease (an inflammation of the blood vessels).
TNF alpha is a pleiotropic molecule that plays a central role in inflammation, immune system development, apoptosis, and lipid metabolism.
